Presentation: Immune Tolerance, Restored – Towards a cure for autoimmune diseases

ABO21009: a first-in-class anti-CD45RC antibody to restore immune tolerance in autoimmune disease

ABO21009 targets CD45RC, which is highly expressed on effector T cells, B cells and plasmablasts but low or absent on regulatory T cells (Tregs) and memory cells. It depletes pathogenic CD45RChigh cells, preserves memory recall responses, and expands Tregs with a suppressive (HOPX-associated) transcriptional signature. In preclinical models it prevented collagen-induced arthritis and abolished autoantibody deposition in APECED. In healthy volunteers (n=24), single ascending doses were well tolerated and confirmed target engagement. Dosing in rheumatoid arthritis patients is ongoing.
